Abstract: | In the beginning of the 20th century, the Russian Nobel prize winner Elie newlineMetchnikoff observed high life expectancy in Bulgarian persons who ate large newlineamounts of fermented milk products. Based on this observation the followed research newlineemphasized, that Lactic acid bacteria plays an important role in retarding colon newlinecarcinogenesis by possibly influencing metabolic, immunologic and protective newlinefunctions in the colon. Various studies have indicated that L. acidophilus MTCC447 newlinemay alleviate retarding colon carcinogen, but ratio of carcinogen neutralisation was newlinediffer for lab condition (in vitro) and intestinal ecosystem (in vivo) is 1: 5 respectively, newlinethe mechanism of intracellular neutralization of carcinogen is also unknown. The aim newlineof the study was (1) To study the anti-carcinogenicity activity of L. acidophilus newlineMTCC 447 and L. acidophilus M, (2) To study the role of abiotic stress (related to newlinehuman intestine) such as Temperature (35 to 60º C) pH (2 to 9), salinity (0.1 to 0.8m newlineNaCl) on carcinogenic protection activity of L. acidophilus MTCC 447 and newlineL. acidophilus M, (3) To study the role of plasmid on anti-carcinogenic activity in newlinestress environment, (4) To study role of stress protein from L. acidophilus MTCC447 newlineon anticarcinogenic activity. L. acidophillus MTCC447 were assayed to determine newlinetheir potential effects on two reference genotoxins: the 4-NQO, which produces DNA newlineadducts and the MNNG, a DNA alkylation agent. Antigenotoxicity was studied both newlinein prokaryotic and eukaryotic cells, using the SOS-Chromotest, a primary DNA newlinedamage assay on Escherichia coli and the Comet assay, for analyzing DNA breakage newlinein enterocytes. newline |
