Abstract: Rice is one of the highly consumed staple foods across the globe, andthe increasing global population is exerting tremendous pressure for sustainable rice production. Besides increase the actual yield through genetic gain, prevention of the losses associated with biotic and abiotic stresses shall also be reduced to sustain the high rice yield. Among the other diseases, rice blast disease by M. oryzae is of a significant threat to global rice productivity. newlineThere are several reports showing the importance of hybrid proline-rich proteins (HyPRPs) involved in various biological functions including plant defence system. In the present study, we identified the genetic makeup of HyPRPsgenes in the rice genome, using in silico analysis. The RNA-Seq data and the qRT-PCR based expression analysis of some selected genes allow us to identify the candidate OsHyPRP gene, namely OsHyPRP16as pathogen inducible gene.The transient overexpression in tobacco indicate the possible role of OsHyPRP16 protein in suppressing the PCD caused by co-infiltrated Bax genes. newlineTo study the effect of R genes under the control of pathogen inducible promoter is considered a better way to minimise the negative effects associated with R genes. Therefore in the present study,cis-regulatory elements OsHyPRP16 promoter region was characterized using deletion analysis.Our results indicate OsHyPRP16promoter is induced in response to different stress hormones, abiotic as well as biotic stresses. The D3 fragment of OsHyPRP16 promoter showed highly induced GUS activity after M. oryzae infection. Through this study, we identify and functionally validate the OsHyPRP16 promoter and identify a promoter fragment responsible for expression under various stresses, including M. oryzae infection.Hence, the OsHyPRP16 promoter identified here could be effectively used for the better management of rice blast disease. newline newline
