Abstract: The oral bioavailability of aliskiren hemifumarate is low due to poor absorption of the drug. newlineThe obstacle may be overcome by designing different systems for the delivery of drug. The newlinedrugs can be delivered by an alternative pathway that is transdermal route newlineThe present work was aimed at preparation of transdermal patches by solvent casting method newlineusing a varying concentration of polymers, i.e., methocel (K15 and K100), ethocel (4 and 10), newlinegelatin, chitosan, eudragit (RL and RS grades) using plasticizer (glycerin and propylene newlineglycol). The release of the final formulation were enhanced by using natural and synthetic newlinepermeation enhancers like limonene, cineol, and#946;-cyclodextrin and oleic acid. As different newlinepenetration acts differently with polymers. newlineThe ratio of drug to polymers and plasticizer was varied and the effect of formulation newlinevariables was studied. Prepared transdermal patches were evaluated for physicochemical newlineproperties, invitro permeation studies, content uniformity, primary skin irritation studies and newlineFT-IR studies. newlineTransdermal patches were prepared by the solvent evaporation technique. The controlled newlinerelease polymers were used for the preparation of patches. The patches were prepared with newlinedifferent polymers and different plasticizer. The drug and polymer interaction study was newlineperformed by Fourier transform infrared spectra. Invitro permeation studies were conducted newlineusing pretreated cellophane membrane using Franz diffusion cell. newlineThe formulated transdermal patch using methocel K 100 M showed good physical properties. newlineThe average weight of patches prepared using glycerin as a plasticizer was ranged from newline42.33-67.00 mg and propylene glycol as a plasticizer were ranged from 40.67-67.67 mg. The newlinepercentage moisture absorption varies between 1.76 to 10.73 for patches formulated using newlineglycerin and 2.28 to 7.97 for propylene glycol patches.
