Abstract: | The present study was carried out to find the antioxidant peptides from the protein hydrolysates of two marine cephalopod mantles, cuttlefish (Sepia brevimana), and squid (Loligo duvauceli) using three different enzymes (trypsin, and#945;-chymotrypsin and pepsin) for 12 hours hydrolysis. All the hydrolysates were evaluated by DH used with OPA method and antioxidant assays. Among these, 7th hour of trypsin hydrolysate of cuttlefish mantle (THC), and 8th hour of and#945;-chymotrypsin hydrolysate of squid mantle (CHS) showed maximum radical scavenging activities. Those selected hydrolysates were purified by chromatographic techniques using FPLC, in ion exchange (IEC) method both the hydrolysates were separated into four fractions. Among these, fractions C from THC and CHS showed maximum antioxidant activity than the others. Furthermore, the active fractions from IEC were again separated by gel filtration chromatography (GFC), where it showed four different fractions from C of THC, and three from C of CHS. After collecting the fractions, the III fraction of THC (DPPH- 38.81 ± 1.07%; RPA- 0.478 ± 0.03 and OH- 40.39 ± 1.27%) and CHS (DPPH- 36.96 ± 1.79%; RPA- 0.3046 ± 0.01 and OH- 34.86 ± 0.83%) were confirmed with the highest activity using UV-visible spectrophotometer newline |
