Abstract: | In the present study, we have reported synthesis of carboxymethyl xanthan gum-graftpoly newlineacrylamide (CMXG-g-PAAm) by using microwave assisted and conventional graft newlinecopolymerization method using ammonium persulphate as a free radical initiator. newlineCMXG-g-PAAm was synthesized by derivatization of xanthan gum (XG) to newlinecarboxymethyl xanthan gum (CMXG) followed by graft copolymerization with newlineacrylamide (AAm), to overcome the limitations of XG (poor solubility in polar solvents newlineand susceptibility to microbial contamination on storage). XG was carboxymethylated by newlineconventional and microwave assisted method. The Synthesized CMXG was further newlinegrafted with AAm, using conventional free radical polymerization and microwave newlineassisted methods. The synthesis process was optimized to yield maximum % grafting and newline% grafting efficiency. The graft copolymers have been characterized by 1H NMR, FTIR newlinespectroscopy, X-ray diffraction measurement, thermal analysis, viscosity measurement newlineand scanning electron microscopy. Further to confirm the safety and applicability of newlinesynthesized polymer, toxicity study, antimicrobial study and drug release characteristics newlinefrom matrix tablets prepared from direct compression and wet granulation methods were newlineexamined. The results revealed that, CMXG was successfully synthesized by an newlineinnovative MW assisted technique, which is eco-friendly and more suitable for mass newlineproduction on commercial basis. Graft copolymers of acrylamide with carboxymethyl newlinexanthan gum were effectively synthesized by the conventional and microwave assisted newlinemethod. Using microwave method, % grafting has been achieved maximum than newlineconventional method even at 40 times less concentration of APS, with same AAm/CMXG ratio. The spectroscopic data confirm that the grafting of acrylamide might newlinehave taken place at -OH group of CMXG, which is supported by a tentative mechanism newlinesuggested for grafting.
