Abstract: Human Erythropoietin EPO is a sialoglycoprotein hormone newlineresponsible for the production of red blood cells Glycosylation has been newlinefound to be imperative for its erythropoietic function whereas asialo EPO and newlineother modified forms are found to have neuroprotective function Due to newlineminute amounts of EPO in blood recombinant methods are used to produce newlineEPO drug EPO traditionally being produced in mammalian systems is newlineessential for treatment of anemia On the other hand aglycosylated EPO newlineproduced using other expression systems can be used for neuroprotection newlineThis study focuses on efficient strategies to produce EPO in E coli and newlinemammalian systems newlineObjective 1 was directed towards overexpression studies of EPO in newlinethe cytoplasm of E coli Effects of downstream box on the expression levels newlineof mature EPO were studied For this a total of nine constructs were made newlineBased on the results an optimal downstream box for the expression of EPO newlinecomprised of expression suppressive codons followed by CAT repeats was newlinederived newline newline
