Abstract: newlineBackground: As per traditional medicine, leaves of plants Premna latifolia Roxb. and newlineCanthium parviflorum Lam. are used to cure the diabetes. newlineObjectives: The present research work focus to assess the effect of ethanolic extract newlinePremna latifolia (EEPL) and Canthium parviflorum (EECP) leaves on metabolism of newlinecarbohydrate and lipid in diabetes and Diabetic Neuropathy (DN) animal models newlinefollowed by fractionation of most promising extract. newlineMethodology: Streptozotocin (STZ) (60mg/kg, i.p.) administered to make animals newlinediabetic. EEPL and EECP were given at dose 100 and 200mg/kg, p.o. as a treatment newlinedaily once for 21 days. Various parameters like body weight, diabetes specific markers newline(insulin and blood glucose) and lipids (triglycerides, total cholesterol, low density newlinelipoprotein and high density lipoprotein) were measured. Enzyme activities concerned in newlinelipid and carbohydrate metabolism namely Phosphogluco-isomerase, Glucose-6- newlinephosphatase, Amylase, Fructose-1,6-diphosphatase, Aldolase, Glucose-6-phosphate newlinedehydrogenase, and Lipase were estimated. Histopathological studies of pancreas were newlineperformed. Further, neuroprotective potential of EEPL and EECP were studied in rats by newlineSTZ induced DN model. DN was assessed in rats by employing hot-plate, tail flick tests newlineand also by measuring levels of Cytokines (namely interleukins and tumor necrosis newlinefactor) also nerve growth factors in sciatic nerves. Oxidative stress also assessed by newlinetesting activity of Thiobarbituric acid reactive substances, Catalase, Reduced glutathione, newlineGlutathione reductase, Superoxide dismutase and Glutathione peroxidase in the sciatic newlinenerves. newlineXVIII newline newlineResults: EEPL and EECP regularizes the serum glucose, lipid profile, insulin, newlinecarbohydrate and lipid metabolizing enzymes level by showing its effect on pancreatic newlineand liver diabetic animals. Extracts increase the pain threshold by normalizing the levels newlineof various Cytokines, improve nerve growth factors and provide protection against newlineoxidative stress. newlineConclusion:
