Abstract: | A multiple drug resistant (MDR) bacteria are a class of bacteria which is resistant to different classes of antibiotics and cause severe complications in humans as well as animals. Such an organism Pseudomonas aeruginosa was isolated from hospital environment. Various biochemical tests were performed to identify the bacteria. The rRNA analysis had identified the bacteria at species level. Drug resistance with respect to different classes of antibiotics was tested. It was found that bacteria were resistant to Ampicillin, Chloramphenicol, Bacitracin, Erythromycin, Gentamycin, Tetracyclin, Polymyxin B and Ciprofloxacin and sensitive to Amikacin, Ceptazidine and intermediate to Ceftriaxone, Gentamycin, Cefoperazone antibiotics. The resistant bacteria were treated with Dimethyl sulfoxide (DMSO) to observe for the sensitivity. It was observed that DMSO resumed the sensitivity of bacteria towards the antibiotics. DMSO had synergistic effect on antibiotic resistant Pseudomonas bacteria especially with respect to the antibiotics such as ampicillin, chloramphenicol, erythromycin, and tetracycline. It was also observed that in presence of 0.5% DMSO the percentage of protein concentration produced by the resistant bacteria was decreased by 20% and 40% decrease was observed in presence of 1% DMSO. Further the SDS-PAGE analysis revealed the appearance of protein bands at 97.4KDa in antibiotic resistant Pseudomonas but the same disappeared completely when treated with DMSO. newline |
