Abstract: | The present work undertaken for Ph.D. is focused on exploration of certain natural metabolites of microbial as well as plant origin which have anti-oxidant as well as anti-cancer activity. The main concept behind this study was to find safer cytotoxics with additional anti-oxidant properties. newlineIn first part of research, anti-oxidant activity of leaf extracts of Azadirachta indica and Calotropis procera and microbial metabolites of Bacillus megaterium and Escherichia coli were assessed by DPPH activity, total anti-oxidant activity and total reductive capability. Microbial metabolite production was also enhanced by media optimisation and varying certain physiological parameters like pH and temperature whereas plant metabolites were extracted by soaking the leaf powder in several solvents (Methanol, Ethanol, Di-Methyl Sulphoxide, Chloroform, Distilled Water, Boiled Extracts and Ethyl Acetate) and analyse its anti-oxidant and anti-cancer potential. newlineCancer is known to be a multifacet disorder with highest mortality rate in the world. Amongst them, ovarian cancer is reported to have highest death rate amongst all gynaecological malignancies. All traditional chemotherapeutic agents generate free radicals and also suffer from Multi Drug Resistance. Anti-oxidants can prove to be a boon for overcoming various cellular damages as they remove free radicals. newlineThe second part of present research was to synthesise silver nanoparticles (AgNp) through greener routes in presence of suitable plant and microbial metabolites and assess the anti-oxidant and anti-cancerous activity of synthesised AgNp. Anti cancer activity of the metabolites as well as AgNp was evaluated by MTT assay against cancer cell line SKOV3 and healthy cell lines HEK 293. newlineComparative analysis showed that microbial metabolites rich in antioxidant potential were capable of synthesising AgNP with better anti-oxidant potential and were also comparatively safer in comparison to the crude extracts of microbial as well as plants with respect to anti-cancer activity on |
