dc.description.abstractThe present study aimed to assess anticancerous potential of metabiotic extract obtained from indigenous probiotic employing colon cancer cell lines and experimental colon cancer model. It was observed that metabiotic extract of Lactobacillus rhamnosus MD14 exhibited potent antigenotoxicity, cytotoxicity and arrested cell cycle by modulating the expression of various oncogenes in colon cancer cells. Further, it was observed that daily administration of metabiotic extract to sprague dawley rats orally along with weekly intraperitoneal injection of 1,2-dimethylhydrazine up to six weeks attenuated early colon carcinogenesis. Metabiotic extract successfully maintained growth rate, acidic fecal pH, lactobacilli count and reduced the level of pro-carcinogenic fecal enzymes, aberrant crypt foci, oxidants, proto-oncogenes and up regulated tumor suppressor gene more effectively with almost normal colonic histo-architecture compared with DMH-treated animals. In totality, the study provided scientific basis for the application of metabiotics independent of probiotics particularly for individuals susceptible to colon cancer.
