dc.description.abstractLipases triacylglycerol hydrolases EC 3113 catalyze the newlinehydrolysis of triglycerides to free fatty acids and glycerol Extracellular lipase newlinesecreted by the gramnegative bacterium Pseudomonas aeruginosa is a newlinepromising candidate for biotechnological applications and is extensively used newlinefor chiral production of racemic compounds newlineIn the first part of this thesis the production of lipase by newlinePseudomonas aeruginosa PGSL 03 isolated from surfactant gel utilizing newlinevegetable oil seeds was investigated Maximum lipase activity was observed newlineat 30C and pH 75 in sesame meal at 2 wv concentration The amount of newlinelipase correlated well with the relative percentage of calcium and C18 n fatty newlineacid esters present in the media The study therefore identifies vegetable oil newlineseeds as a simple and economical lipidrich media viable at an industrial newlinescale Ammonium Sulphate precipitation of culture supernatant resulted in newlinepartial purification of the enzyme 428 fold with 906 yield newline newlineen_US
