dc.description.abstractAsthma is a chronic inflammatory disease of the airway, characterized by bronchial hyper responsiveness, mucus hyper production and airway obstruction. These pathological changes are directly associated with the infiltration of inflammatory cells and release of cytokines. The plant Myxopyrum serratulum (M. serratulum) is an important medicinal plant traditionally used for the treatment of asthma, cough and nerve complaints. Although the extract of M. serratulum has several pharmacological properties, the evaluation of bronchodilatory and anti-asthmatic effect has not been reported yet. Hence, the present study was designed to evaluate the bronchodilatory effect of the extract using guinea-pig model and inhibitory effects on ovalbumin-induced airway inflammation in a mouse model of allergic asthma. Further, we evaluated the suppressive effect of the extract on lipopolysaccharide-induced inflammation in RAW 264.7 murine macrophages. Additionally, the isolation of active compounds from M. serratulum was performed by preparative HPLC analysis and identified using spectral studies, such as IR, 1H NMR, 13C NMR, 2D Cosy and Mass spectrophotometer. The active compound of MEMS was identified as Myxopyroside and the anti-inflammatory effect of Myxopyroside was studied in LPS-induced inflammation in RAW 264.7 murine macrophages. The protective effect of MEMS and Myxopyroside on the production of pro-inflammatory mediators like nitric oxide (NO) and reactive oxygen species (ROS) and pro-inflammatory cytokines (IL-1and#945;, IL-1and#946;, IL-2, IL-4, IL-6, IL-10, IL-12, IL-17A, IFN-and#947;, TNF-and#945;, G-CSF and GM-CSF) in LPS-stimulated RAW 264.7 murine macrophages were also studied. newline newline newline
