Abstract: | Neuroprotection is often defined as preventing neuronal death or restoration of neuronal functions or number by a phytochemical or synthetic molecule Parkinsons disease is a progressive nigro striatal neurodegenerative disorder including the clinical signs of Parkinsonism Among various vulnerable factors affecting nigro striatal tract excitotoxicity is one of the important pathological mechanisms Cyclic guanosine monophosphate cGMP pathway is one of the causal factor for excitotoxicity leading to degeneration apoptosis of nigro striatal neurons The hypothesis of the study was stated as Besides antioxidant property anti apoptotic property of MCE provides neuroprotection by impeding MPTP toxicity induced apoptosis The Momordica charantia extract was prepared and analysed with LCMS This extract was screed for antiparkinsonian properties in in-vitro model of SHSY5Y cells with MTT assay and TBARS assay The in-vivo study included was sub acute MPTP C57BL6J mice model and behavioural biochemical histopathological evaluations were done LCMS analysis revealed presence of 193 molar mass invitro study revealed cytoprotective doses of MCE invivo study revealed that MCE attenuated MPTP toxicity and retained SOD levels GSH levels decreased NO levels dopamine This inhibition of MPTP toxicity by MCE was also seen in behavioral parameters and immuno histochemical histopathological evaluations The MCE synthesised with water extraction and alcohol precipitation method showed antioxidant and neuroprotective activity in both invitro human neuroblastoma SHSY5Y cells MPP model and invivo MPTP mice model of parkinsons disease Further studies and applicable drug formulations can be made for prophylaxis and treatment of Parkinsons disease newline newline newline |
