Title: Formulation and Evaluation of Nanoparticles for Improved Therapeutic Activity of Drugs Used for Neurodegenerative Disorders

Abstract: Tacrine hydrochloride loaded polymeric nanoparticles were designed, optimized and prepared by newlineemulsion polymerization method. Three widely accepted and physiologically safe polymers newlinePLGA, PLA, and MPEG-PCL were used to for the development of nanoformulations.The newlinephysiochemical characterization and other performances of the formulations were evaluated and newlinesuitable for treatment of Alzheimer s disease. In this study, Tacrine hydrochloride is selected as a newlinemodel drug. The prepared formulations were found to be stable at room temperature for period of newline1 year. The nanoformulations are intended for sustained release of the drug for a period of 24 newlinehours and this may reduce the dosing frequency, thereby minimizing the side effects. The in vitro newlinecell line was carried out on SH-SY5Y cell line suggests reduced toxicity to normal cells and newlineincreased cellular uptake. The pharmacokinetic studies reveal that the transport of more amount newlineof the TH across the BBB cause improvement in bioavailability and increase the effectiveness of newlineAlzheimer s disease. Thus, the nanoparticles of TH can potentially improve the therapeutic newlineactivity of tacrine, with simultaneous reduction in decrease of free drug in general systemic newlinecirculation. This will considerably reduce the adverse drug reactions of TH, it should be noted newlinethat the predominant adverse drug reactions over the therapeutic activity is the major reason for newlineto withdraw in the market. Hence the nanoparticular system of TH indicates the possibilities to newlinebring back the drug to the market. However more patient studies are suggested. newline
