Abstract: | Lipase producing bacterial strains were screened from soil samples of edible oil processing industries. Out of 92 bacterial isolates, 26 strains (AKL01 AKL 26) were found to be positive for lipase production on Tributyrinagar medium. Further screening on phenol red olive oil agar and rhodamine olive oil agar medium revealed that the isolate AKL 13 as best lipase producer. Based on various cultural, morphological, biochemical and molecular characterization, isolate AKL 13 was identified as Bacillus subtilis AKL 13.The newly isolated Bacillus subtilis AKL 13 strain and its 16S rDNA sequence are submitted to Microbial Culture Collection, National Centre for Cell Science, Pune (Accession no. MCC 3235) and GeneBank, NCBI (Accession no. JX092100), respectively. Screening of the appropriate variable for optimization of the lipase production medium was performed using one variable at a time approach followed by response surface methodology. Among various parameters tested in optimization studies, the medium containing glucose (0.33 % w/v), used palm oil (0.75 % v/v) and Triton X100 (0.015 % v/v) yielded maximum lipase production (4286 U/mg of protein) and biomass (2.38 g/L). B.subtilis AKL 13 showed 0.435 h-1 specific growth rate in optimized medium. Further, maximum lipase production was recorded at 14th h of cultivation period. newline |
