dc.description.abstractIn vivo anticancer activity of Cayratia trifolia (L.) were studied in MNU and newlinetestosterone induced prostate cancer rats. After the induction of prostate cancer, newlinebiochemical parameters such as total protein, enzymatic and non enzymatic antioxidants, membrane bound enzymes were decreased whereas LPO, tumor markers, nucleic acid constituents were increased in prostate gland and seminal vesicle tissues. The hematological, lipid parameters were altered and prostate markers, renal markers and liver markers newlinewere increased in serum. Those levels were brought back to near normal upon treatment newlinewith stem ethanolic extract of Cayratia trifolia (L.) and standard drug finasteride. No significant changes were observed on treatment with plant extract alone group which indicated that there is no toxic substance present in Cayratia trifolia (L.) which was confirmed by the histopathological analysis of organs liver, kidney, prostate gland and seminal vesicle. With chromatographic and spectroscopic techniques, the natural compounds such as sitosterol-1-acetyl propionate, eicosenoic acid, 1-heptacosanol and 1-pentacosanol were newlineisolated and identified from the stem part of ethanolic extract of Cayratia trifolia (L.). Among the four, sitosterol-1-acetyl propionate is the newly isolated compound from Cayratia trifolia. Therefore sitosterol-1-acetyl propionate was chosen for further in silico and in vitro analysis. The molecular docking studies revealed that, the isolated compound of newlinesitosterol-1-acetyl propionate have good inhibitory activity against PTEN, NKX3.1 and 5-and#946; reductase enzyme when compared with finasteride (FDA approved drug for prostate cancer) and ADME properties prediction results of the compound under acceptable range. Although the isolated compound sitosterol-1-acetyl propionate have good cell growth inhibitory activity at minimal concentration (IC 50) by the MTT assay.
