Title: Pharmacological Evaluation of Agents Altering Insulin Metabolism in Experimentally Induced Diabetes in Rats

Abstract: newline Objective: To partially purify the Insulin Degrading Enzyme (IDE) from rat kidneys newlineand to estimate the insulin in the IDE assay. To purify the plant protease inhibitors newline(PPI`s) for in-vitro evaluation of inhibitors against the Insulin degrading enzyme and newlineselected PPI`s will be screened for in-vivo anti-diabetic activity in streptozocin newlineinduced diabetes in rats. newlineMethod: The IDE was partially purified from wistar rats and plant protease inhibitors newlinewere also partially purified from the various plant sources. The RP- HPLC method newlinewas developed and standardized for estimating remaining insulin in the assay. In-vitro newlineIDE assay was performed by incubating insulin, IDE and various plant protease newlineinhibitors. The acute toxicity study was performed by administering 2000mg/kg in newlinerats. The selected inhibitors were administered orally at a dose level of 100 mg/kg and newline200 mg/kg body weight and sub-acute toxicity for fifteen days. In the long term newlinetreatment of Streptozocin induced diabetic rats; the degree of protection was newlinedetermined by measuring insulin levels, blood glucose levels, biochemical and newlinehistopathology studies newlineResult: SCPI, MCPI at a dose of 200 mg/kg body weight for 28 days, suppressed the newlineelevated blood glucose and lipid levels in Streptozocin induce diabetic rats. newlineConclusion: Our findings indicate the SCPI and MCPI, suppressed the elevated blood newlineglucose and lipid levels in Streptozocin induced diabetic rats. newlineKeywords: Plant Protease inhibitors; hypoglycemic effect; Diabetes mellitus; Insulin newlineDegrading Enzyme; IDE.
