Abstract: Systematics, phylogenetics and taxonomy are the scientific fields of species newlinediscovery, delimitation, description, classification and evolutionary history. With newlineincreasing extinction threats to thousands of living species, a reliable and consistent newlineway of identifying and classifying species for conservation is of utmost importance. newlineUsing cytochrome oxidase subunit I gene (COI) gene as a marker, this study reflected newlinethe phylogenetic relations of twelve gastropod species collected from East Cost of newlineIndia. Our taxonomic sampling of the chosen gastropod partial COI DNA sequences newlinewas run in Basic Local Alignment Search Tool (BLAST) to find the available closest newlineand most similar sequences of species in terms of percent base pair similarity, was newlineanalyzed using the Maximum likelihood (ML) method. newlineThe data were investigated priori to cladogram reconstruction in order to find newlinethe most appropriate dataset for reconstructing gastropod phylogeny. The chosen 61 newlinegastropod nucleotide sequences of partial COI gene were analyzed to evaluate for newlinenucleotide substitution saturation using DAMBE software. It was revealed that values newlineobtained for a critical index of substitution saturation were less than the values shown newlinein the index of substitution saturation. The related t-value at the degree of freedom newlinewere found to be statistically significant indicating that there was little saturation at all newlinethe three base positions of codons suggesting the scope for substitutions. Further, it newlinewas meant to estimate the substitutions within purines and pyrimidines (Transitions) newlineand between purines to pyrimidine (Transversions). It was shown that the total newlineestimate of 48.75 transitions and 51.26 transversions must have occurred in all 61 newlinegastropods chosen in the present study. Interesting perception was seen concerning the newlinepreferred-codon usage. The values signifying effective number of codons (ENC) range from 38.6 to 55.9 among 44 out of 61 genera indicated the preference towards a few codons for expression.
