Abstract: Polycystic ovary syndrome (PCOS), the most common endocrine disorder affecting women of reproductive age is associated with state of low grade chronic inflammation and depends on multiple genetic and environmental factors. Intercellular adhesion molecule-1 (ICAM-1) is an important biomarker of inflammation and endothelial dysfunction.Elevated levels of inflammatory markers including ICAM-1 are demonstrated in women with PCOS suggesting increased cardiovascular risk in these women.Recent evidence indicates a significant linkage between chromosome 19p13 loci and multifactorial diseases that have an inflammatory involvement.ICAM-1 gene located on chromosome 19p13 has genetic variations which are common in populations and many studies have described associations between these polymorphisms and inflammatory disease. Therefore, the polymorphism of ICAM-1 gene represents a strong positional and biological candidate for the susceptibility to the development of inflammatory and metabolic diseases which includes PCOS. In the present study the possible role of K469E polymorphism on exon 6 and G241R polymorphism on exon 4 of ICAM -1 gene in development of PCOS among Kashmiri (North Indian) women has been examined. newlineOral contraceptive pills (OCP s), are the usual first choice of treatment, after weight reduction and lifestyle modifications in women with PCOS, as they ameliorates hyperandrogenic symptoms. Because OCP s are known to alter coagulation and fibrinolytic markers towards a more hypercoagulable state and epidemiological studies have shown increased risk for venous thromboembolism among users of OCP s compared to non- user. Therefore treatment with OCP s may worsen the already deranged inflammatory and coagulation parameters in this population. In this study effect of administration of OCP s (0.03mg- ethinyl-estradiol and 0.15mg- levonogestrel) on mRNA expression of ICAM-1 gene in women with PCOS was examined.
