dc.description.abstractBackground: Inflammatory diseases are still one of the most important heath problems in the world. A large number of disorders associated with inflammation. Currently used non steroidal anti-inflammatory drugs have severe side effects. Therefore there is a need for new safe approaches for such diseases that is the use of natural botanical sources. newlineAim: To evaluate anti-inflammatory potential of leaves of Ipomoea aquatica Forsk and fruits of Vernonia anthelmintica Willd along determination of possible mechanism of action, characterization of more active constituent and development of effective formulation. newlineMaterials and Methods: The authentified leaves of I.aquatica and fruits of V.anthelmintica were subjected to pharmaconostical study and phytochemical study. Estimation of total phenolics and total flavonoids were done with hydro alcoholic extract of I.aquatica leaves (HAEIA) and hydro alcoholic extract of V.anthelmintica fruits (HAEVA). The anti inflammatory activity of HAEIA and HAEVA was evaluated by formalin-induced rat paw oedema, serotonininduced rat paw oedema, antihistamine activity, prostaglandin synthesis inhibition activity, membrane stabilizing activity, protein denaturation inhibition activity and proteinase inhibitory activity. By using fractionation and preparative TLC, two compounds (IA-I and IA-II) were isolated from HAEIA. newlineHPTLC chromatography used to quantify isolated compound IA-I. The 32 Full Factorial design was employed to evaluate main effects and interaction effects of IA-I and IA-II on the carrageenan induced rat paw oedema model for anti-inflammatory study in order to optimize the combination. The best combination was used to develop topical gel formulation. The prepared topical gel was subjected to anti-inflammatory study and physical and stability evaluation. The characterization of more effective isolated compound IA-I was done with help of its physical properties, chemical tests, TLC, UV, FT-IR, Mass spectroscopy and 1H NMR spectroscopy. newline
