Abstract: | newlineBackground: Preservation of primary tooth in a disinfected state is the best space newlinemaintainer. Pulp therapy is the ideal treatment alternative but it has its own challenges. newlineSuccessful pulp therapy in primary teeth depends on reduction and elimination of root newlinecanal infection; although no techniques for root canal instrumentation have been newlinefollowed. Intracanal medicaments are essential to reduce these virulent and pathogenic newlinebacteria for successful pulp therapy. newlineObjectives: newlinePrimary objective: newline1. To test the antimicrobial activity of various combinations of intracanal newlinemedicaments and vehicles on selected pathogens from deciduous molars with newlinenecrotic pulp. newlineSecondary objective: newline2. To identify selected micro-organisms from root canals of infected deciduous newlinemolars by means of microbial culture. (Streptococcus spp, Enterococcus newlinefaecalis and Porphyromonas gingivalis) newline3. To detect difficult to grow microorganisms from root canals of infected newlinedeciduous molars by means of Polymerase Chain Reaction. (Enterococcus newlinefaecalis, Porphyromonas gingivalis and Treponema denticola) newlineMethodology: The Phase 1 consisted of in vitro bactericidal activity of vehicles using newlinebroth dilution method followed by in vitro antibacterial activity of endodontic newlinemedicaments and vehicle combinations using agar well diffusion against ATCC strains newlinewas done using pure and commercial drugs. newlineXXV newline newline newlineThe Phase 2 consisted of the main study, assessment of the antibacterial activity newlineof the three intracanal medicaments, ie DAP, modified-DAP and Chlorhexidine with newlinethree vehicles, namely Glycerine, PEG and PG against three selected endodontic newlinemicroorganisms using an Ex-vivo model.
