Title: Development and Evaluation of Natural Polymeric Film Loaded with Rivastigmine and Donepezil for Transcranial Drug Delivery

Abstract: The present aim of research work was to develop and evaluate rivastigmine and donepezil loaded natural polymeric film for transcranial drug delivery. The natural polymers used were collected from various natural sources and was isolated by simple economic method. The biomaterial recovered from the concentrate was subjected for various physicochemical properties. Polymeric nature was confirmed by means of IR Mass NMR. Two drugs i.e. rivastigmine and donepezil and ten natural polymers were utilized individually at different ratio to prepared hundred nano formulations by modified nanoprecipitation method. Prepared batches were subjected for various evaluation studies like particle size zeta potential SEM TEM surface entrapment etc. On the basis of several evaluation parameters like particle size zeta potential PDI drug entrapment surface entrapment in vitro diffusion profile as well as surface morphology two formulations RM5 and FD4 were selected as optimized formulation and subjected for further evaluation study. Result of ex vivo release study of optimized formulation suggested that prepared formulations were able to release the drug at a same rate and extent in biological condition also. In vivo pharmacological screening of AD on animal model was indicating that optimized formulation act effectively when administered via transcranial route. In vivo release study conclude that the drug effectively reach into the brain via transcranial route and provide a concentration that required to produce desired therapeutic effect. In vivo Brain Cholinesterase estimation study revealed that transcranially applied optimized formulations significantly reduce brain cholinesterase activity in animals when compared with scopolamine treated group. From the above experimental study it can be concluded that rivastigmine and donepezil loaded natural polymeric film can effectively treat Alzheimer disease and improve the patient compliance. newline newline newline newline newline newline
