dc.description.abstractquotObjective: 5-ASA and sodium salicylate loaded liposomes incorporated into PEG-4-acrylate and CCRGGC could probably suit the needs for developing a disease responsive drug delivery system which will serve as a prophylactic therapy for asthmatic patients. newlineMethodology: The two drug candidates were subjected to formulation development by preparing liposomes and encapsulating with PEG-4- acrylate microgels and subsequent characterization and in vitro studies. The developed formulations were attributed to MTT assay, Apoptosis study, western blot analysis of Iand#954;B kinase and p38-and#947; along with ELISA (TNF-alpha, IL-6, IL-13), nitrate and nitrite, myeloperoxidase (MPO) and malonyldialdehyde (MDA) levels at three dose levels in lipopolysaccharide (LPS) induced RAW 264.7 cells followed by in vivo evaluation using Ova induced allergic asthma in Balb/C mice at three dose levels. The Pharmacokinetic and bio distribution study was also performed. newlineResults: The in vitro study results revealed significant dose dependent reduction in the levels of the aforementioned parameters accompanied by the reduced expression of Iand#954;B kinase and p38-and#947; in cells treated with 5 ASA and SS microgels when compared to LPS treated cells. The in vivo studies further justified the in vitro findings except for the leucocyte count which was significantly increased compared to Ova control. The histology studies have shown promising airway structural restoration at highest tested dose levels of both the formulations. The formulations also have shown localization in the lungs for 20 hr in pharmacokinetic evaluation, justifying the selection of the formulation design. newlineConclusion: The developed formulations have revealed potential findings both in vitro and in vivo against inflammatory and oxidative stress parameters involved in allergic asthma. Overall, the liposomes encapsulated microgels of 5-ASA and SS have revealed prospective findings in this study against allergic asthma and can serve as potential candidates for treating the same.
