Abstract: The study was aimed at finding out the prevalence of Candida tropicalis and C.auris in candidaemia condition in our tertiary care centre and finding out the mechanisms by which the organisms resist the activity of fluconazole In the study we found that C.tropicalis was the predominant species causing candidaemia and resistance to fluconazole was found in nearly 11percent of the isolates The mechanism of fluconazole resistance in C.tropicalis was studied by PCR amplification and sequencing the erg 11 gene in fluconazole resistant and susceptible isolates and analyzing them for presence of mutations The expression of various genes like erg 11 mdr1 and cdr 1 was also studied by reverse transcriptase quantitative PCR and was found out that erg11 and mdr 1 were over expressed in fluconazole resistant C.tropicalis than fluconazole susceptible C.tropicalis The effect of fluconazole on the gene expression was also studied This gave the importance of mutation and gene expression in the resistance of C.tropicalis Whole genome sequence analysis was done to study the genetic basis of fluconazole resistance in Cauris The erg 11 gene sequence was analyzed for single nucleotide polymorphisms also, the 3D structure of Erg 11 was predicted and its interaction with fluconazole was studied This gave an insight into the interaction of fluconazole and Erg 11 newline
