Abstract: | Peanut bud necrosis disease caused peanut bud necrosis virus (PBNV), an enveloped newlineRNA virus is a very serious problem in peanut or groundnut (Arachis hypogaea L.) newlineproduction. Thrips Frankliniella fusca and Frankliniella occidentalis, are the vectors newlinefor the virus, present a difficult and complicated challenge from the perspective of newlineboth epidemiology and disease management. Simply controlling the vector typically newlinehas not resulted in control of bud necrosis. A genetic engineering approach is proved newlineto be of great potential for resistance enhancement against PBNV. In the present study newlinetransgenic peanut lines with resistance to PBNV were developed from de embryonated newlinecotyledon and immature leaf explants of groundnut cultivars Kadiri-6 (K-6) and K-134 newline(Vemana) using Agrobacterium tumefaciens strain LBA 4404 harboring Nucleocapsid newlineProtein gene of Bud necrosis virus driven by CaMV 35S promoter. From the cultivar newlineK-6, 1612 shoots were regenerated from a total of 327 de-embryonated cotyledon newlineexplants. The survival of rooted and hardened plants in glasshouse was 92% and final newlinerecovery of putative transgenics was 42.4%. In case of cultivar K-134, 69 deembryonated newlineexplants were co-cultured and 398 shoots were regenerated. Survival of newlineplants in the glasshouse was 89% and the putative transgenic recovery was 27.6%. A newlinetotal of 3822 shoot buds were regenerated from 697 immature leaf explants from the newlineseeds of cultivar K-6 were developed. Survival of plants under glasshouse conditions newlinewas 91% and a 13.9% of final recovery of putative transgenics was observed. |
