Title: DEVELOPMENT AND EVALUATION OF ACECLOFENAC LOADED MUCOADHESIVE MICROPARTICLES

Abstract: The present study was aimed to develop and evaluate the mucoadhesive newlinemicrocapsules of Aceclofenac. Aceclofenac is a non-steroidal anti-inflammatory drug newlineused for relief of pain and inflammation in osteoarthritis, rheumatoid arthritis and newlineankylosing spondylitis. It has a short biological half life of 3-4 h. The conventional newlineAceclofenac tablets administered 2 to 3 times a day due to its short biological half life newlineproduces side effects like gastric ulceration and gastric bleeding. The side effects of newlineconventional Aceclofenac tablet have been attenuated by designing the drug in the form newlineof mucoadhesive microcapsules. The mucoadhesive microcapsules of Aceclofenac were newlineformulated by orifice ionic gelation method and emulsification ionic gelation method newlineemploying polymers like Hydroxypropyl methyl cellulose, Sodium carboxy methyl newlinecellulose, Methyl cellulose and Carbopol along with Sodium alginate in the drug polymer newlineratio 1:1 and 1:2. newlineFT-IR study was carried out to determine any drug polymer interaction. The study newlinerevealed that there was no interaction between the polymers and drug. Prepared newlinemicrocapsules were evaluated for particle size, SEM analysis, percentage yield, flow newlineproperties, drug content, drug entrapment efficiency, percentage moisture loss, swelling newlineproperty, in vitro drug release, drug release kinetics and in vitro wash-off test. The newlinemicrocapsules should exhibit good flow property during filling into the capsules. So the newlineformulations were evaluated for micromeritic parameters and the values were found to be newlinewithin the prescribed limits for all formulations. newlineThe microcapsules were discrete, spherical and free flowing. Average particle size newlinewas found in the range of 922.3±2.36and#956;m to 977.5±5.21and#956;m in orifice ionic gelation newlinemethod and 736.4±3.36and#956;m to 862.7±5.71and#956;m in emulsification ionic gelation method. newlineLow coefficient of variation (lt 2.0%) in percentage drug content of all formulations newlineindicates uniform distribution of drug throughout the microcapsules.
