Abstract: | newlineOral submucous fibrosis OSMF is a potentially malignant disorder and a fibrotic condition caused due to arecanut chewing OSMF lays its foundation on oxidative stress inflammation and inflammation mediated fibrogenesis The study aimed at evaluating the antioxidant anti inflammatory and antifibrotic activity of three medicinal plants Centella asiatica L LECA Ocimum basilicum L LEOB and Origanum vulgare L subsp hirtum LEOV and their active compounds asiatic acid linalool and thymol in an arecoline inducible human buccal fibroblast HBF cell line model In the present study the ethanolic LECA LEOB and LEOV were prepared via cold maceration technique LEOV exhibited maximum activity via the TAO and RP assay LECA via the NO and SO radical scavenging assay and LEOB via the DPPH assay at 1000 µg/mL Arecoline treatment upregulated proinflammatory IL 1B IL 6 and TNFand fibrotic TGFB1 COL1A2 and COL3A1 cytokines simultaneously downregulating IL 10 in cultured HBFs at higher concentrations of 25 µg/mL and 50 µg/mL On pretreatment with LECA LEOB and LEOV downregulated the elevated mRNA expression of proinflammatory and fibrotic cytokines upregulating IL 10 at higher concentrations of 62 5 µg/mL and 125 µg/mL The same was observed with pure compounds at concentrations 7 8 to 3125 µgmL |
