Abstract: The present work is devoted to the development of standard conditions and methods for the analysis of antiepileptics, antidepressants, fluoroquinolones, quinolones, and acyl homoserine lactones using preconcentration techniques like solid phase extraction (SPE) and microextraction in packed syringe (MEPS) followed by the separation with high performance liquid chromatography (HPLC) with ultraviolet visible (UV) detection and gas chromatography-mass spectrometry (GC-MS) from various environmental and biological samples. Along with this during method development on these instruments the physical parameters which are the characteristics of the separation of these compounds used are also considered for the study. Various parameters like retention factor (k), separation factor (and#945;), and efficiency factor (N) were optimised to describe the resolution equation which predicts the effect of variations in these factors in controlling resolution within the chromatographic column. The chapter wise summary of the work done is presented below. CHAPTER 1: General Introduction This chapter deals with general introduction to drugs and pharmaceutical compounds selected for the study and techniques used for the analysis of these compounds in the proposed work. CHAPTER 2: Review of Literature This chapter provides an overview of different applications of SPE, MEPS, HPLC and GC-MS for the analysis of drugs chosen for the study reported in literature. CHAPTER 3: Analysis of Antiepileptic Drugs using MEPS-HPLC-UV/GC-MS Systems In this chapter HPLC-UV and GC-MS determination of the antiepileptics in human plasma and urine samples using MEPS as a sample preparation technique is discussed. CHAPTER 4: Analysis of Tricyclic and Nontricyclic Antidepressants by MEPS-LC- UV/GC-MS In this chapter, analysis of tricyclic and nontricyclic antidepressants by MEPS-LC-UV/GC-MS are conferred and compared. This chapter deals with the sensitive and accurate quantitative determination methods for five antidepressants i.e. amitriptyline, imipramine
