Abstract: Background: Doxorubicin hydrochloride (Dox/DOX) is a well-known semisynthetic, newlineanthracycline antibiotic used in cancer chemotherapy but has some severe side effects newlinenamely cardiotoxicity and bone marrow toxicity. Regardless of these widespread claims newlineof Dox in cancer management, its use is constrained owing to the most serious side effect newlinelike cardiomyopathy, subsequently leading to congestive heart failure. Nanoparticles can newlinebe employed for targeting anticancer drugs in which it is anticipated that nanocarriers like newlineGNPs would deliver the drug at the unhealthy cancerous tissues via two mechanisms, i.e. newlinepassive or active mechanism. Even though several nano-medicines have been formulated newlineto improve the anticancer effect of Dox, most of the investigations are restricted only to newlinebrief in vitro anticancer activity. Several polymeric Dox formulations display poor newlinebiodegradability, biocompatibility and face stability concerns and poor industrial newlinescalability. Hence, green synthesized GNPs could serve as a potential carrier wit h an newlineadvantage of improved biodegradability, biocompatibility, improved loading capacity, newlinetargeting ability, scalability and stability. newline newlineAim and Objectives: The aim of the present research was to investigate the effect and newlinetargeting potential of Dox loaded gold nanoparticles (D-GNPs) formulation in the newlinetreatment of chemically induced fibrosarcoma bearing mice. Hence the objectives of the newlinestudy are newline newline newline1. Synthesis of GNPs by green chemistry method and its characterization. newline2. Loading and characterization of doxorubicin onto synthesized gold nanoparticles. newline3. To study anticancer activity of doxorubicin loaded GNPs and to generate correlation newline newline newlinewith associated side effects. newlineMethodology: GNPS were synthesized by using PM and NK extract and purified by high newlinespeed centrifuge at 17000 rpm at 4ºC for 20 min. Formation of GNPs was initially newlineobserved by visual color change and confirmed by UV-Visible spectroscopy. Antioxidant newlineassay (TAC, TFC, TPC) of plant extract before and
