Abstract: Proteases, the biocatalysts with ability to catalyze cleavage of peptide bonds are most studied among the hydrolytic enzymes. Alkaline proteases are one very unique class of proteases with heightened physiological and commercial significance. Alkaline Proteases have multitude of applications in various industrial sectors such as detergents, leather processing, food, pharmaceutical, effluent treatment etc. Besides, alkaline proteases also play significant role in the fields of organic synthesis, diagnostics and protein engineering. Moreover microbial proteases hold back a huge share of 60% from the world wide enzyme market. Recent years have witnessed a growing realization over issues pertinent to environment across the world. This intensified interest in enzymes and white biotechnology to develop eco-friendly products and processes. newlineThe present thesis work has been scripted in 4 chapters of which the first chapter comprises of a detailed introduction of enzymes and their physiological and commercial significance. An introduction to the protease, a class of enzymes belonging to hydrolases family is presented. This chapter has been concluded with a note on Response surfacemethodology, an efficient statistical method to optimize the process parameters. The second chapter includes review of literature wherein an in detail survey of literature pertaining to alkaline proteases is portrayed. This chapter particularly staged the multidimensional survey on alkaline proteases which briefly include: alkaline protease producing organisms, assay methods, fermentative production (Submerged and Solid state fermentations) of alkaline proteases, characterization studies conducted on alkaline proteases and applications. Precisely, the present study has been initiated with objectives to isolate, purify and characterize an alkaline protease from a soil organism. Primarily soil samples were processed to isolate a soil bacterium with ability to produce an alkaline protease enzyme.
