dc.description.abstractThe thesis embodies the synthesis and characterization of new metal-based antitumor agents. In-vitro DNA binding studies of the metal-based chemical entities that can be utilized to treat chronic diseases in particular cancer/malignancies were carried out employing various bio-physical techniques. These studies reveal that these complexes can act as potential chemotherapeutic agents, and show promise to overcome inherent drug resistance and may exhibit fewer side effects. The thesis is divided in six chapters which includes Chapter I Introduction giving a detail overview of literature relevant to this field and also helps to justify the objectives of the work done. Chapter II is experimental where all details of methods and physical measurements employed in this work are given. Chapter III-VI embodies the work done i.e. de-novo design, synthesis of novel metal-based chemotherapeutic drug agents and exploring their in-vitro binding propensity towards the specific target DNA at the molecular level. Chapter III deals with the synthesis of new heterobimetallic complexes [C16H22ON5Cl4CuSn]Cl and [C16H22ON5Cl4NiSn]Cl derived from R(+)-phenylglycine chloride hydrochloride and dichlorodimethyl bis(4-pyrazole N2) tin (IV). These complexes were thoroughly characterized by spectroscopic (IR, 1H, 13C, 119 Sn NMR, XRD, EPR, UV-vis, ESI-MS) analytical methods and TEM and AFM visualization techniques. In the complexes, the geometry of copper and nickel ions were square pyramidal while tin ions were present in hexacoordinate environment. Various biophysical methods viz. electronic absorption titrations, fluorescence and cyclic voltammetric of free complex and in presence of DNA were carried out. DNA condensation is an essential process to transport a therapeutic vector to its moleculartarget. It includes the collapse of extended DNA chain into compact orderly particles containing one or more molecules.en_US
